Hello,

I am a high school senior with a Senior Thesis focused in Biochemistry. I am working with simple Peptide Nucleic Acid monomers and protecting groups to test if the protecting groups could allow synthesis and self assembly of PNA oligomers. I know you are involved in both Open Gel Box and OpenPCR, so I was wondering if you know if PNA would function the same way DNA should in both of these.

By the way, I recommend there by an OpenHPLC at some point so I can hack together an analytic biochemistry lab.

Thanks,

Out of interest, how would someone get hold the primers for a PCR? I would love you guys to get an open gene machine going!

Cheers.

I saw the video,
http://www.youtube.com/watch?v=zrejvnVfZJE

and I would love to play with it! I'm only a sophomore in high school, but I love chemistry and biology and pretty much hacking in general!

What I'm curious though, is that with 3 billion base pairs of dna, which or how much can the device look at?

Thanks!

Can you just use a standard digital camera to take a picture of the gel through the orange filter? Has anybody tried this? Also, any idea if this system would work with "GelRed" http://www.biotium.com/product/price_and_info.asp?item=41003&Sub_Section=09A . Excitation and Emission spectra are shown in link.

Hey, I like this project, I just wanted to say Congrats and Good Luck!

I'll talk about it in my lab, it looks like a real cheap alternative to the systems we're using now (gel box + camera + computer); The only thing is SYBR safe is more expensive than Ethidium bromide although less dangerous (even though also being a DNA intercalant)... Can the blue leds also reveal BET stained gel ? I think its absorption spectra is in the UV as well as 450-500nm so it should be fine.
Anyway, keep up this great initiative, and all the others more and more needed in this politic-economic context where only very high and excellent achievers in research are rewarded with funds, selecting research economically viable on short term. Your initiative will help not only "biohackers" to do it themselves but also small labs struggling financially!

Cheers,

Hey Guys,

Absolutely fabulous product. I've been thinking about making my own but didn't know where to start. Do you guys have a parts list and instructions. I'm trying to figure out how to go from the files in the hardware section to an actual product. I mostly need the transilluminator as I already have a gel box. But what do I do with the pdf schematic? Where do I go to buy the parts, etc.

Can someone help a newbie out???

Thanks,

Hi,

My gelbox is on it's way! I'm hurriedly ordering some of the SYBRsafe dye as well.

I usually use EtBr, though, and to be fair will probably still find a way to do so.

I've got a couple of questions regarding the manufacture of the gelbox...

1) what is the top filter? Is it comparable to the Cokin orange 002A that I would normally filter with?

2) is there anything in the clear base that would block large portions of UV in the 312 nm range? See any problem with just sticking a UV lamp under the base rather than the blue LEDs?

Thanks!

--Derek

At first I would like to say CONGRATULATIONS! You've made a really great work!
At second, I want to propose an improvement to the website. I think you should include some pictures with results of an experiment, just to have an idea about how is the fluorescence of the DNA bands. Maybe with another with a hand near or something that allow to know how big it's really, because in Europe, we are not familiarized whit your metric system.

Thank you very much, Carlos Toscano from Seville (Spain)

Hi again! I would like to know how works (if it works) using the gel box with SYBR green I & II and SYBR gold. Also EtBr. Because I know that absortion spectrum for all the SYBR members if very near. And with the BrEt I think that is not going to work but maybe yes or something (I don't know the spectrum for the BrEt).

Thanks, Carlos Toscano

Biotech newbie here. Am looking for a way to do my own Hep C and HIV tests on used and discarded needles and other drug paraphrenalia left in our neighbourhood. Wondering if Pearl gel box will help me in accomplishing this. If not, any pointers? Thanks.
